Clinical Relevance
Coagulation factor II, or prothrombin, is a Vitamin K-dependent Glycoprotein encoded by the F2 gene on chromosome 11. Activated thrombin enzyme is crucial in hemostasis and thrombosis. It converts fibrinogen to fibrin for blood clot formation, stimulates platelet aggregation, and activates coagulation factors V, VIII, and XIII. Thrombin also inhibits coagulation by activating protein C.
Prothrombin g20210a mutation involves the substitution of adenine for guanine at position 20210 within the 3' untranslated region of the prothrombin gene. Individuals with this variation have a predisposition to developing abnormal clots in either veins (deep vein thrombosis or DVT) or lungs (pulmonary embolism or PE). The mutation is associated with an increased risk of myocardial infarction and susceptibility to recurrent miscarriages.
