NucleoDx® Tris-Acetate EDTA (TAE) buffer is a widely used buffer in molecular biology experiments because it provides optimal conditions for several important processes. The buffer's composition makes it ideal for a variety of applications. Tris serves as the primary buffer and helps to maintain the pH level of the solution. Acetate assists Tris in this regard and also acts as a source of ions to conduct the electrical current in electrophoresis experiments. EDTA plays a critical role by chelating magnesium ions that can inhibit the enzymes used in the experiment. The buffer pH is approximately 8.4 at room temperature.
TAE Buffer is suitable for various molecular biology applications, including agarose and polyacrylamide gel electrophoresis, DNA and RNA gel staining , maintain a constant pH for separation of molecules and DNA sequencing. TAE buffer is generally preferred due to its lower ionic strength, which helps to reduce the interference of ions on the separation process.
